PITTSBURGH, June 2006 � Investigational oral growth hormone secretagogues could emerge as the fountain of youth for aging baby boomers. Two measures of physical function in men and women 65 or older were improved by 10% after 12 months of treatment with one of the growth hormone secretagogues.

After six months of treatment with the growth hormone secretagogue, there was a statistically significant improvement in tandem walking (P=0.02) and after 12 months there was a significant improvement in stair walking (P=0.04), Dr. Merriam and colleagues reported at the International Congress of Neuroendocrinology.

Patients receiving active treatment also reported that they felt more vim and vigor when taking oral growth hormone secretagogues. They regretted stopping HGH at the end of the study.

Tandem walking involves one foot in front of the other, a test motorists are asked to do to test sobriety, and tests both physical control and function, Dr. Merriam said. Stair climbing is a standard test of physical function and one of several measures used to evaluate the ability to live independently. That benefit, he said, would be extending the period of fit aging so that people could live independently longer.

The study enrolled 395 men and women ages 65 to 84, who were living independently but who had mild functional limitations such as reduced grip strength or reduced walking speed. Compared with placebo, all active doses "prompted acute growth hormone peak and an increase in overnight growth hormone secretion."

"All doses of growth hormone secretagogue were associated with an increase in lean body mass," Dr. Merriam said.

Dr. Merriman said growth hormone production is virtually non-existent in older people and the lack of the hormone is thought to contribute to loss of lean muscle mass, which contributes to decreased function. Early in the 1990s researchers began studying growth hormone injections as a treatment to slow or reverse aging. Those studies confirmed that injection could increase circulating growth hormone and improve lean muscle mass, but results were mixed on improving function.

Dr. Merriman said that he and others believe that growth hormone secretagogues offer a more natural approach to increasing growth hormone since these compounds trigger increased production of growth hormone in "brief pulses which better mimic the body's own production of growth hormone."

This study was supported by Pfizer, which is developing capromorelin, the ghrelin-mimetic used in the study. Dr. Merriam said that Merck has a similar compound in development.
